肥胖大鼠肝脏、骨骼肌叉头状因子O1基因表达对胰岛素抵抗的影响

Effect of FoxO1 expressions of mRNA and protein on insulin resistance in liver and muscle tissues of obese rats

摘要 目的观察肥胖大鼠肝脏和骨骼肌叉头状因子O1(FoxO1)的表达,探讨FoxO1在肥胖和胰岛素抵抗发生中的作用。方法 30只SD大鼠随机分为对照组(NC,常规饲料)和高脂组(HF,高脂饲料)。饲养10周后测定有关指标。结果 10周后与NC组相比,HF组体重、血糖、胰岛素、TG、TC、腹内脂肪/体重均升高,肝脏和骨骼肌FoxO1mRNA表达量分别增加84%和88%(P<0.01)。TG、HO-MA-IR及腹内脂肪/体重是肝脏FoxO1mRNA表达的独立相关因素,HOMA-IR及腹内脂肪/体重是骨骼肌FoxO1表达水平的主要影响因素。结论肥胖大鼠肝脏和骨骼肌FoxO1表达量升高,可能是肥胖状态下引发胰岛素抵抗的机制之一。 Objective To investigate the effect of FoxO1 expressions of mRNA and protein on insulin resistance. Methods 30 SD rats were randomly divided into 2 groups: control (NC), high-fat diet (HF). The parameters were detected after feeding for 10 weeks. Results The body weight (BW), visceral fat/body weight ratio (VF/BW), FPG, Ins, TG, TC, and HOMA-IR index were significantly higher in HF group than in NC group. The mRNA levels of FoxO1 in liver and muscle of HF group were increased by 84 % and 88% respectively compared with NC group (P〈0.01). TG, HOMA-IR and VF/ BW were independent factors influencing FoxO1 mRNA in liver(r^2 =0. 141, 0. 271, 0. 273, all P〈0. 01), while HOMA-IR and VF/BW were independent variables of muscle FoxO1 mRNA expression(r^2 =0. 460, 0. 219, P〈0. 01). Condusions The increased expression of FoxO1 in high-fat-fed rats is related to obesity and insulin resistance.
