摘要 目的:通过减少瞬目频率建立蒸发过强型兔干眼模型。方法:新西兰大白兔12只随机分为实验组A,B和对照组C,每组4只。两实验组模拟荧屏终端(video display termi-nals,VDT)操作者的瞬目特点,用铁夹固定兔眼睑将其瞬目频率减少为实验前的1/3,8h/d。造模前和造模后3,7,10,14d行Schirmer试验、角膜荧光素染色和泪膜破裂时间(break-uptime,BUT)检查,造模前后自身对照。对照组置于相同环境下不进行干预。14d处死A,C组兔取结膜、角膜和泪腺组织行光学显微镜观察及杯状细胞计数对照;B组停止干预,于17,21,28d观察指标,28d处死取相同组织行光学显微镜观察及杯状细胞计数。结果:除A组7d和B组3d外,两实验组其余时间点Schirmer试验值与实验前相比都无明显改变(P>0.05)。从3d起,两实验组的角膜荧光素染色评分显著升高,BUT显著减少(P<0.05),随着时间推移变化程度逐渐加重。14dB组停止干预后上述差异仍存在。光镜下两实验组的角结膜上皮层以增生性改变为主,泪腺组织无明显变化。14dA组4个方位球结膜的杯状细胞数量都比对照组C组少,差异显著(P<0.01),以鼻侧和颞侧球结膜差别更明显。28dB组各方位球结膜杯状细胞数量仍明显低于对照组C组(P<0.01)。结论:减少瞬目频率能成功地建立蒸发过强型兔干眼模型。 AIM : To establish a rabbit model of evaporative dry eye by decreasing blink rate. METHODS- Twelve New Zealand white rabbits were randomly divided into 2 experimental groups (A and B) and 1 control group (C) , with 4 rabbits in each group. Simulated the blink characteristic of video display terminal(VDT) operators, rabbits of experimental groups were fixed eyelids with clamps and decreased blink rate by one third of before for 8 hours everyday. Rabbits of experimental groups were fixed eyelids with clamps and decreased blink rate by one third of before, simulated the characteristic of blink of VDT syndrome,which lasted for 14 days and 8 hours everyday. Schirmer test, fluorescence staining and tear film break-up time (BUT) were performed before experiment and on 3, 7, 10, and 14 days, with own control. Control group were in the same environment without intervention. Conjunctiva, cornea and lacrimal glands tissues of rabbits in group A and C were removed for histopathological observation under light microscope and goblet cell number was counted after 14 days, with control in two groups. Then experimental intervention discontinued in group B and the former tests were still taken on 17, 21, and 28 days, as well as histopathological observation after 28 days. RESULTS: Schirmer tests were no significant difference( P 〉 0. 05) after experiment compared with baseline except on day 7 in group A and on day 3 in group C. From the day 3 on, the level of fluorescence staining significantly increased and BUT scores were significantly decreased (P 〈 0.05) in two experimental groups, and then the difference became more and more prominent. Without intervention from day 14, the difference still existed in group B. Histopathological observation revealed that proliferation mostly occurred in cornea and conjunctiva epithelium and no change was observed in lacrimal glands tissues. Conjunctival goblet cell density in group A was significantly lower ( P〈 0.01 ) on every side on day 14 than group C and was most prominent on the nasal and temporal sides. On day 28 goblet cell density in group B was still lower significantly than group C (P〈0.01). CONCLUSION, Rabbit model of evaporative dry eye could be established by reducing blink rate successfully.
