Cu,Zn,Se,Cd离子对河流弧菌Ⅱ毒性的研究

摘要 本文用10倍稀释法确定了河流弧菌Ⅱ在Cu,Zn,Se,Cd离子中的半致死浓度是3.56×10-2、4.46×10-3、3.69、2.78×10-5μg/ml。细菌吸收了这几种离子后其超微结构发生了明显变化。这些离子能诱导细菌内的核糖体增多,DNA纤丝凝聚成棍棒或块状。以Cu、Zn离子诱导DNA纤丝聚集较为明显,类核区扩大或出现透明区,而Se、Cd离子处理的菌核区不明显。而且4种离子对细胞壁也有不同程度的破坏,Zn离子使细胞壁粗糙与胞膜分离明显,Cu、Zn离子处理的样品细胞质集中在周边,细胞质内出现明显的电子致密颗粒。尤其是4种离子混合作用的细菌,上述现象更为明显。Se离子毒性相对较弱,细菌的超微结构没有明显的变化。Cd离子处理的细菌超微结构变化明显。首先,细胞壁呈泡状外突,最后小泡处破裂。细胞质内出现许多电子密度大的颗粒(可能是聚缩的DNA),类核区不明显,但范围比对照正常菌明显扩大,其内的DNA纤丝聚缩成短棍状或块状,散布在细胞质中。 This paper reported semilethal concentration of Cu,Zn,Se and Cd ion to Vibrio fluvialis Ⅱby ten times dilution method,it was found to be 356×10-2、446×103、369or278×105μg/ml respectively.After the bacteria absorb Cu,Zn,Se and Cd,their ultrastructures were changed.These ions could induce the increase or bacteria ribosome,the DNA aggregated to stick of cube form.The Cu,Zn ion induced DNA fibril clearer than Se,Cd ion induced.The Cu,Zn ions also induced nucleoid area expanded or transparent area appeared,while the transparent area did not appeared with Se,Cd treated.Four ions could destory their cell wall.Zn ion coarsened cell wall and made cell membrane seperated from the wall.The cytoplasma of Cu and Zn ions treated bacteria concentrated on periphery.If a mixture of Cu,Zn,Se and Cd ions treated the bacteria the phenomenon became clearer.Toxicity of Se ion was weak,bacteria ultrastructure did not change.Cd ion changed bacteria ultrastructure prominently.First,cell wall presented bubble protruding,later bubble bursted out.There were a lot of high density electron particle in bacteria cytoplasm,nucleoid area was not clear.DNA fibril aggregated to stick or cube Which dispersed in cytoplasm.
