摘要
采用两种方法分离山羊卵巢腔前卵泡。方法1(M-1):将卵巢剪成碎块(<3mm),用01%胶原酶消化处理,吸管吹打,离心洗去胶原酶并置入网筛内过筛后检卵。方法2(M-2):用组织切碎机将卵巢切成碎块,经吸管吹打制成的组织悬液置入网筛内过筛后检卵。将不同年龄山羊对称分成两组,M-1和M-2分离的腔前卵泡头均数分别为1164和1383个,其中正常卵泡比例分别为296%和274%,差异不显著(p>005)。采用M-1分离初情期前山羊腔前卵泡效果最佳,平均每只羊可获得16082个腔前卵泡,其中正常卵泡数平均为4936个;而在成年山羊则分别为3500和667个(p<001)。采用M-2分离成年山羊腔前卵泡头,其效果明显优于M-1(p<005)。
Two methods are used for isolation of preantral follicles from goat ovaries. Method 1 (M-1): Ovaries are cut into small pieces (<3mm) with the scissors, the fragments are then treated with 01% collagenase solution, pipetting. The suspension is centrifuged and the preantral follicles are sorted. Method 2 (M-2): Ovaries are cut into small pieces with a tissue chopper. The suspension is pipetted and filtered successively through 450 and 200μm mesh filters. Goats with different ages are divided into two groups. M-1 and M-2 result in 1164 and 1383 preantral follicles per pair of ovaries, among which the percentage of normal follicles are 296% and 274% respectively (p>005). M-1 is available for the isolation of preantral follicles from prepubertal goat ovaries, resultion in 16082 preantral follicles per pair of ovaries, with 4936 normal follicles. However, the isolating efficiency of M-1 used in adult goat is significantly decreased, resulting in only 3500 preantral follicles and 667 normal follicles per goat (p<001). M-2 is superior to M-1 in the isolation of preantral follicles from adult goat ovaries (p<005).
