约氏疟原虫感染早期易感和抵抗小鼠树突状细胞亚群和表型的变化特点 被引量:1

DENDRITIC CELL SUBPOPULATION AND PHENOTYPE CHARACTERISTICS OF SUSCEPTIBLE AND RESISTANT MICE DURING THE EARLY PHASE OF PLASMODIUM YOELII INFECTION

摘要 为探讨致死型约氏疟原虫(Plasmodium yoelü 17XL,P.y 17XL)感染早期,树突状细胞(Dendritic cells,DCs)亚群和表型的变化特点,本试验利用P.Y17XL感染易感的BALB/c和抵抗的DBA/2小鼠,制备感染前和感染后第3、5天小鼠脾细胞悬液,采用流式细胞分析技术检测2种小鼠脾细胞悬液中髓样Dcs和浆样DCs的数量以及表面表达MHCⅡ类分子和CD80分子的DCs的百分含量。结果表明,BALB/c小鼠和DBA/2小鼠分别呈现以浆样DCs和髓样DCs为主的增殖模式;2种小鼠表面表达MHCⅡ类分子的Dcs的数量均于感染后第3天开始明显升高,在第5天达到最高水平,但BALB/c小鼠的表达水平明显低于DBA/2小鼠;DBA/2小鼠表面表达CD80分子的DCs的数量在感染后第3~5天呈逐渐增高趋势,其水平明显高于BALB/c小鼠。而BALB/c小鼠表面表达CD80分子的DCs的数量仅在感染后第5天出现有意义的升高。结果显示,P.y 17XL感染早期,BALB/c和DBA/2小鼠在DCs亚群的增殖模式、成熟表型特征性分子的表达水平和时相上均存在显著差异。 To investigate the dendritic cell (DC) subpopulation and phenotype characteristics during early infection of P/asmod/um yoelii ( P. y) 17XL. Susceptible BALB/c and resistant DBA/2 mice were infected by i. p. injection of P. y 17XL parasitized erythrocytes. Mice from normal and infected groups were sacrificed on 0, 3rd and 5th day post infection for collection of spleen cells. Flow cytometry was applied to quantitatively analyze myeloid DCs, plasmacytoid DCs and DCs expressing MHC H or CD80. It was found that BALB/c mice and DBA/2 mice respectively showed plasmacytoid DC and myeloid DC predominant proliferation model. The population of DCs expressing MHC II significantly increased on day 3 and peaked on day 5 post infection in BALB/c and DBM2 mice. However, the levels of BALB/c mice were less than those of DBM2 mice. Meanwhile, the population of DCs expressing CD80 in DBM2 mice showed gradually increasing trend from day 3 to day 5 after infection, which were higher than those in BALB/e mice. Nevertheless, the percentage of DCs expressing CDSO in BALB/c mice significantly increased only on day 5 post infection. These results indicated that significant differences existed in DC subpopulation proliferation model, and in levels and phase of characteristic molecules reflecting mature phenotype in BALB/c and DBA/2 mice during the early stage of P. y 17XL infection.
