摘要
为研究燕麦β-葡聚糖对高脂血症大鼠胆固醇代谢的影响,将58只雄性SD大鼠随机分为基础组(n=10,饲喂基础饲料)和模型组(n=48)。模型组饲喂高脂饲料3周后,将造模成功的高脂血症大鼠随机分为高脂对照组、低剂量组、中剂量组和高剂量组,每组10只,分别饲喂含0、6、12、24g/kgβ-葡聚糖高脂饲料。在试验进行到2、4和6周时测定大鼠的血浆总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C);在试验末期测定血浆载脂蛋白B(ApoB)和AI(ApoAI)、大鼠肝胆固醇、肝微粒体胆固醇含量、肝脏7-α羟化酶活性、粪胆固醇、粪胆汁酸和肠胆汁酸含量。结果表明,不同剂量的β-葡聚糖都极显著地降低空腹血浆TC、LDL-C和ApoB水平(p<0.01),而对HDL-C和ApoAI无显著影响。不同剂量的β-葡聚糖都显著降低肝胆固醇(p<0.05),增加微粒体胆固醇含量和7-α羟化酶活性(p<0.05),促进肠道、粪便胆汁酸和胆固醇排泄(p<0.01)。燕麦β-葡聚糖可有效降低血脂,抑制肝脏胆固醇沉积,提高7-α羟化酶活性,利于胆固醇和胆汁酸的排出,其作用效果具有剂量和时间依赖性。
In order to sudy the effects of oat β-glucan on cholesterol metabolism in experimental hyplipidemic rats, Fifty-eight male SD rats were randomly divided into two groups:control group(n=10) fed basal diets, model group(n=48) fed high-fat diets for 3 weeks. Hyplipidemic rats were divided into 4 treatments (n=10): high lipid control treatment, low-dose, medium-dose and high-dose oat β-glucan treatments, which fed with high-fat diets plus different contents(0, 6,12 and 24g) β-glucan/kg. The content of TC, LDL-C, HDL-C level in plasma were measured respectively at the end of weeks 2,4 and 6. ApoAI and apoB in plasma, hepatic cholesterol, microsome cholesterol and 7α-hydroxylase (CYP7A1) activity, fecal cholesterol and bile acid concentrations were determined at the end of the experiment. The resuhs showed that the contents of TC, LDL-C and apoB in plasma were significantly decreased and had no effects on the contents of HDL-C, apoAI of the hyplipidemic rats by three different oat β-glucan groups comparing to high lipid control group. The groups of differents doses of β-glucan significantly reduced hepatic cholesterol concentrations (p〈0.05), increased hepatic microsome cholesterol contents and 7α-hydroxylase (CYPTA1) activity significantly (p〈0.01) and further increased the bile acids excretion and cholesterol concentrations (p〈0.01). The results suggested that oat β-glucan could reduce blood lipid, control accumulation of hepatic cholesterol, increase 7α-hydroxylase activity, the bile acids and cholesterol excretion. Obvious dose and time-dependence was observed.
