摘要
建立了一种灵敏的测定血清促红细胞生成素（ＥＰＯ）的ＥＬＩＳＡ夹心法。该法应用人重组促红细胞生成素（ｒｈＥＰＯ）免疫家兔获得的，并经亲和层析纯化的多克隆抗ＥＰＯ，与各种血液组份和细胞因子无交叉反应性。灵敏度０．４ｍＵ／ｍｌ～０．５ｍＵ／ｍｌ。批内变异２％～８％，批间变异８．９％～２５．１％。血清与标准剂量反应曲线平行。与ＲＩＡ法测定结果相关良好（ｒ＝０．９４）。
A sensitive sandwich enzymelinked immunosorbant assay for erythropoietin(EPO)was developed using a polyclonal antibody raised in rabbits against human recombinant EPO(rhEPO)and purified over a rhEPO affinity chromatography column. No reaction was obtained with a variety of blood components and cytokines. The sensitivity was 04～05 mU/ml. The intraassay variation ranged from 2%～8%, while the interassay variation ranged from 89%～251%. Serum doseresponse curve were paralled to the standard. A good correlation between the RIA and ELISA data was observed(r=094).
