摘要
生长激素刺激培养新生大鼠心肌细胞,用含MBP凝胶分离法测定细胞外信号调节酶(ERK)活性。用WesternBlooting法检测磷酸化的p38MAPK,观察心肌细胞内MAPK超家族活性变化,探讨心肌细胞GH信号传导途径。结果发现:生长激素激活心肌细胞ERKs呈时间和浓度依赖性关系。而对p38MAPK无磷酸化激活作用。表明心肌生长激素信号传导通过MAPK超家族成员ERK途径,而与p38MAPK途径无关。生长激素诱导的心肌细胞ERK激活,可能是生长激素心脏效应的重要机制之一。
The cultured cardiac myocytes of neonatal rats were stimulated by growth hormone (GH),and then the activity of extracellular signal-regulated kinase(ERK) was assayed with the method of MBP-containing gel and the phosphorylated p38MAPK was detected with Western Blotting.The changes of MAPK superfamily activities were observed to explore GH signal transduction pathway in cardiac myocytes.The results showed that GH activated ERKs in cardiac myocytes in a time- and a dose-dependent manners, however,GH did not cause phospho-activation of p38MAPK in cardiac myocytes.This study indicates that GH signal transduction in cardiac myocytes is closely related to ERK,a member of MAPK superfamily,but is not relevant to p38MAPK pathway.GH-induced ERK activation in cardiac myocytes may be one of the important mechanisms by which GH affects the heart.
