难治性颞叶内侧型癫痫患者脑组织内BDNF及其受体TrkB的表达 被引量:2

Expression of BDNF and its receptor TrkB in brain tissue resected from patients with intractable mesial temporal lobe epilepsy

摘要 目的研究难治性颞叶内侧型癫痫(MTLE)患者脑组织内脑源性神经营养因子(brain derived neurotrophic factor,BDNF)及其受体TrkB的表达,探讨其在癫痫发病机制中的作用。方法采集11例难治性MTLE患者手术中切除的颞叶和海马脑组织,用免疫组化、逆转录-聚合酶链反应(RT-PCR)法检测BDNF、TrkB的蛋白和mRNA表达,并与非癫痫患者对照。结果与对照组相同部位比较,患者组颞叶和海马中的BDNF蛋白和TrkB蛋白的表达明显增加(P<0.01),BDNF mRNA和TrkB mRNA在患者组颞叶(P<0.01)和海马(P<0.05)的表达明显增加。结论难治性MTLE患者颞叶和海马中BDNF和TrkB受体表达增高,可能在癫痫发生、发展中具有重要作用。 Objective To study the expression of brain derived neurotrophic factor (BDNF) and its receptor TrkB in the brain tissue resected from patients with intractable mesial temporal lobe epilepsy (MTLE) and discuss their role in the pathogenesis. Methods Immunohistochemistry and reverse transcription polymerase chain reaction (RT-PCR) were used to test the expression of BDNF and TrkB in the surgically removed hippocampi and temporal lobe cortex tissues from 11 patients With intractable MTLE, and the results were compared with those of controls. Results Expression of BDNF protein and TrkB protein were significantly increased in temporal lobe cortex and hippocampal tissue resected from patients with intractable MTLE compared with that of controls(P 〈 0. 01 ). Expression of BDNF mRNA and TrkB mRNA were significantly increased in temporal lobe cortex( P 〈 0.01 ) and hippocampal tissue ( P 〈 0.05 ) resected from patients with intractable MTLE compared with that of controls. Conclusion The results indicate that the expression of BDNF and TrkB increases in the temporal lobe and hippocampus of patients with intractable mesial temporal lobe epilepsy, and it may play an important role in the generation and development of epilepsy.
