摘要
本文建立了快速、简便测定人血清中氨氯地平的ＨＰＬＣ法，分析柱为 ＯＤＳ－５Ｓ（ １５０ × ４．０ｍｍ）不锈钢柱，流动相为甲醇：０．０３Ｍ磷酸二氢钾缓冲液（９∶１）的混合液。流速为１．０ｍｌ／ｍｉｎ，以硝苯地平为内标物，检测波长为３６６ｎｍ，蛋白沉淀剂选用１５％高氨酸。氨氯地平测定的线性范围为４～１２μｇ／ｍｌ，ｒ＝０．９９９２，最低检测限度为 ４ｎｇ。日内平均相对回收率为 １０．２４ ± ５．４０％，日间平均相对回收率为 ９９．８７ ± ６．２３％。本方法专一性好，放置２４ｈ对含量测定无影响。
A rapid specific and sensitive HPLC menthod for the determination of amlodipine in serum was developed, using a stainless steel column (ods - 5s, 150 × 4.0mm) with nifedipine as internal standard. The mobile phase was a mixture of methanol - KH_2PO_4 buffer(0.03mol/L), flow rate 1ml/min. Wavelength of the detector was set at 300nm. The linear correlattion was observed within the range of 4 - 12μg/ml(r=0.9992) and detect limitation at 4ng. The mean recoveries of the method were 100.24 ± 5 .4O% (intra - day)and 99. 87 ± 6.23% (inter - day).
