摘要
从发病的暗纹东方鲀体内分离、鉴定了一株致病性嗜水气单胞菌,测定其毒力,命名为NT-1。以NT-1为抗原,免疫28周龄的罗曼蛋鸡,用水稀释脱脂处理,聚乙二醇(PEG)一步法制备高效价的卵黄抗体,微量凝集试验、ELISA检测其效价及特异性;不同浓度卵黄抗体的粗提液与NT-1菌液混合注射ICR小鼠,检测特异性卵黄抗体的抑菌效果。结果表明,微量凝集试验测得的特异性抗体效价达1∶16以上,ELISA效价达1∶12 800以上;高效价、特异性的卵黄抗体可维持90 d,特异性卵黄抗体ELISA效价达1∶640时对感染NT-1的ICR小鼠的保护率达到了100%。
The bacteria strain NT-1 was isolated from Takifugu obscurus and which were identified as Aeromonas hydrophila by bacteriological method. Hens were immunized with the bacteria. One-step polyethylene glycol (PEG-8000) precipitation procedure was used to purify the immunoglobulin Y (IgY). The titres of purified IgY were detected by Microagglutination test and Enzyme-linked Immunosorhent Assay (ELISA). The titer of Microagglutination in the IgY was higher than 1:16. The ELISA titers in the purified IgY was higher than 1:12 800. High titers IgY to Aeromonas hydrophila could be induced by immunization successfully, the specific activity last more than 90 days. When the ELISA titer in the IgY was higher than 1:640, its protection rate to the infected ICR mice was 100%.
