PTTG在肺癌中的表达及其与血管生成的关系 被引量:2

The expression of PTTG in lung cancer and its relation with tumor angiogenesis

摘要 目的探讨垂体瘤转化基因(PTTG)在肺癌组织中的表达,分析其与临床病理各指标的相关性,并探讨其与肿瘤血管生成之间的关系。方法通过免疫组化法检测72例肺癌组织及10例正常肺组织中PTTG的表达,同时应用CD34单克隆抗体标记,测定微血管密度(MVD)。结果①79.2%的肺癌中PTTG表达为阳性,与正常肺组织相比有显著性差异(P<0.05);肺癌组织中MVD评分与正常肺组织相比有显著性差异(P<0.05);②PTTG的表达均与年龄、性别无明显相关性(P>0.05);与肺癌的TNM分期、组织学分级、淋巴结转移显著相关(P<0.05);③PTTG的表达与MVD显著相关(P<0.05)。结论①PTTG的表达及MVD测定可以作为判定肺癌恶性程度和预后的有效生物学指标;②PTTG在导致肿瘤发生的机制中可能起一定作用;③PTTG可能参与血管生成,而肿瘤血管生成则可能是其致癌机制之一。 Objective To study the expression of pituitary (PTTG tumor-transfroming gene)in human lung cancer and determine the relationship of PTTG expression and its relation with tumor angiogenesis. Methods Archival primary lung cancer (n = 72) and normal lung epithelia (n' = 10) were analyzed for PTTG expression by sABC immunohistochemistry using specific monoclonal antibodies. The microvessel density(MVD) was detected using CD34 staining. Results 79.2% (57/72) of lung carcinomas showed positive expression of PTTG, which was significantly different from the negligible expression in normal lung epithelia (P 〈 0.05 ) ; The score of lung carci.nomas was significantly higher than that of normal lung epithelia(P 〈0.05);(2) PTTG expression level was correlated with TNM stage,lymphnode metastasis, histological grade. (3) The score of MVD in PTTG positive lung cancer was significantly higher than that of PTTG negative lung cancer(P 〈 0.05 ). Conclusion (1)PTTG and MVD may be useful markers for reflecting tumor malignant behavior and estimating prognosis of human lung cancer; (2)PTTG may be the most important mediator of the tumorigenic pathway;(3)Tumor angiogenesis may be related to the carcinogeneses and tumor progression of PTTG.
