三康胶囊对缺氧小鼠糖代谢和HPA轴功能的影响 被引量:5

摘要 目的探讨三康胶囊对缺氧小鼠糖代谢和下丘脑-垂体-肾上腺(HPA)轴的影响。方法灌胃给予高、低剂量的三康胶囊(SKC,1.5、0.5g·kg-1),模拟高原缺氧环境建立小鼠缺氧模型,通过糖代谢相关指标(肝糖原、肌糖原、葡萄糖-6-磷酸酶和血糖)、应激反应的相关指标(肾上腺指数、HPA轴功能)以及HPA轴和糖代谢之间的关系,探讨其抗缺氧作用机制。结果与缺氧模型组相比,SKC能增加缺氧状态下小鼠血糖水平、肝脏糖原含量和葡萄糖-6-磷酸酶(G-6-Pase)的活性,同时使肾上腺指数和血浆中促肾上腺皮质激素(ACTH)、皮质酮和胰岛素(Ins)水平升高。结论SKC具有提高小鼠抗缺氧能力的作用,其机制可能与其调节HPA轴功能,增加肝脏中糖原的输出和G-6-Pase的活性,保证机体重要脏器对葡萄糖的需求有关。 Aim To study the anti-hypoxia effect of Sankang Capsule (SKC) in mice and its relationship with the hypothalamic-pituitary-adrenal (HPA) axis and glucose metabolism. Methods SKC was given by i.g. at the dose of 1.5, 0.5g·kg^-1, to the anoxic mice under low pressure conditions similar to the plateau environment. The mechanism of anti-hypoxia effect of SKC was investigated by determining the glucose metabolism index the stress-re- lated index and the relationship between the HPA axis and the glucose metabolism. Results After the administration of SKC, not only the plasma glucose level, the activity of glucose-6-phosphatase and the content of hepatic glycogen, but also the levels of plasma ACTH, corticosterone and insulin were increased compared with the anoxic model group after the administration of the drug. Conclusion SKC could enhance hypoxia tolerance in mice. The mecha- nism may be related to adjustment of the function of HPA axis, increase of the output of hepatic glycogen and the activity of glucose-6-phosphatases and the glucose supply to important organs of the body.
