矮牵牛花药植株再生体系的建立 被引量:4

Establishment of Plant Regeneration from Anther of Petunia Hybrida

摘要 为了提高矮牵牛花药愈伤组织诱导率,推动矮牵牛花药培养的应用研究。对白色、紫色、粉红色、大红色四种基因型矮牵牛花药进行培养,研究了不同浓度激素配比及不同浓度蔗糖、麦芽糖和白砂糖对花药诱导的影响。结果表明,四种基因型采用6-BA和IBA组合诱导效果较好。获得较高诱导率的激素配比依次为:白色MS+6-BA1.0 mg.L-1+IBA 1.5 mg.L-1,紫色MS+6-BA 1.0 mg.L-1+IBA 1.0 mg.L-1,粉红色MS+6-BA 1.5 mg.L-1+IBA 1.5 mg.L-1,大红色MS+6-BA 1.5 mg.L-1+IBA 1.0 mg.L-1。麦芽糖诱导效果好于蔗糖,白砂糖诱导效果明显差于前两者。 In order to improve the Petunia hybrida callus induction rate, and promote Petunia hybrida anther culture of applied research. Anthers of four genotypes of Petunia hybrida were cultured to evaluate the effects of different proportions of hormone and different concentrations of sucrose, maltose and sugar on adventitious shoot induction. The results showed that the medium combined with 6-BA and IBA was the best for callus induction in the four genotypes. The optimal media for the highest induction ratio were as follows: MS +6-BA 1.0mg·L^-1+IBA 1.5mg·L^-1 in white genotype, MS+6-BA 1.0mg·L^-1+IBA 1.0mg·L^-1 in purple genotype, MS+6-BA 1.5 mg·L^-1+IBA 1.5 mg·L^-1 in pink genotype and MS+6-BA 1.5mg·L^-1 +IBA 1.0mg·L^-1 in red genotype. As a carbon source, maltose was more effective than sucrose and sugar on anther culture of Petunia hybrida.
