舒血宁注射液对高碘致培养血管内皮细胞损伤的保护作用 被引量:3

Protective effects of shuxuening injecta on vascular endothelial cell injury in vitro due to iodine excesses

摘要 目的探讨舒血宁注射液对高碘致人脐静脉血管内皮细胞损伤的保护作用。方法体外培养人脐静脉血管内皮细胞株HUECV-304,在培养液中预先加入不同浓度的碘化钾和舒血宁注射液。在碘化钾的最终浓度分别为4.0、5.0、6.0、8.0μmol/L组中分别加入浓度为0、2、5、10μL/mL的舒血宁注射液,观测4,8,12和24 h时细胞形态、细胞计数、MTT法测定细胞活性及上清液中SOD的含量。结果①在碘浓度5.0μmol/L以上组,4 h时可见内皮细胞内有中毒颗粒出现,且随浓度增加和时间延长而增加。加入舒血宁后,碘浓度5.0μmol/L以上组该变化明显减轻,且与时间和剂量相关(P<0.05);②随时间的延长及舒血宁浓度的升高,细胞增殖活性受到抑制的碘浓度相应升高(P<0.05);③SOD含量随碘浓度的升高而升高(P<0.05),加入舒血宁后,SOD含量变化不明显。结论舒血宁注射液对高碘所引起的内皮细胞损伤有一定的对抗作用,其作用存在时间-剂量效应关系;舒血宁的作用可通过增强细胞的抗氧化作用拮抗高碘的损伤作用。 Objective To explore the protective effects of shuxuening injecta on cultured vascular endothelial cell injury due to iodine excesses. Methods HUVEC was cultured to 24 h with iodine (4.0, 5.0, 6.0, and 8.0 μnol/L) and shuxuening injecta (0, 2, 5, and 10 μL/mL). Morphology and number of HUVEC, and the MTT activities and superoxide dismutase(SOD) of the culture medium on 4, 8, 12, and 24 h were determined. Results 1. Poison granules were found in the endothelium of the 5.0 μmol/L groups and increased with an increase of time and dose of iodine. The above was significantly alleviated in the shuxuening groups and decreased with an increase of time and dose of shuxuening( P 〈 0.05). 2. With an increase of dose and over time of shuxuening, the iodine dose to inhibit the cell growth was increased( P 〈 0.05). 3. SOD activity positively correlated with iodine dose and was not changed in the shuxuening group( P 〈 0.05). Conclusion Shuxuening injecta can counteract injury due to iodine excesses in a dose-and time-dependent manner. Shuxuening may enhance cell anti-oxidization so as to reduce injury induced by iodine excess.
