摘要
目的:建立温莪术的双向电泳技术(2-DE)体系。方法:采用TCA/丙酮沉降法提取蛋白,并用超速离心去除杂质。采用固相pH胶条在IPGphor TM Ⅲ等电聚焦系统上进行等电聚焦,采用Bio-Rad电泳仪进行SDS-PAGE电泳,以银染法染色。结果:得到了上百个点的双向电泳图谱。结论:本研究成功建立了温莪术蛋白双向电泳技术体系。
Objective: To establish the system of 2-D gel electrophoresis proteome of the leaf of Curcuma wenyujin. Methods: The total proteins of root were extracted with trichlo- roacetic acid (TCA)/acetone precipitation method and ultracentrifugation,and then separated by two-dimensional electrophoresis (2-DE), which was an IPGphor TM unit with immobile pH gradient strips used in and the Ettan TM Dalt H system in SDS-PAGE. The 2-D gel was stained with silver. Results: Near hundred 2-D electrophorogram was gained by this method. Conclusion: A system of two-dimensional eleatronhoresis of curcuma wenvu.iin is successfully established.
