经诱导健康人与肿瘤患者CIK和DC-CIK对肺癌细胞杀伤作用的比较研究 被引量:5

A Comparison of Killing Effect on Lung Cancer Cell between CIK and DC-CIK induced from Healthy and those from Cancer Patient

摘要 [目的]比较健康人与肿瘤患者来源的CIK和DC-CIK对肺癌细胞的杀伤作用的差异。[方法]分别将健康人和肺癌患者来源的单个核细胞在体外诱导成CIK和DC;分别用A549细胞和肺腺癌原代细胞的肿瘤抗原冲击DC,体外行DC-CIK共培养;流式细胞仪检测两种来源的DC表面CD40、CD80、CD86和HLA-DR的表达和CIK和DC-CIK中CD3+CD56+细胞比率差异;利用LDH释放法测定同来源CIK和DC-CIK在不同效靶比下对A549细胞和肺腺癌原代细胞的杀伤活性。[结果]两种来源的单个核细胞在体外均可诱导生成CIK和成熟的DC;健康人来源的DC表面CD40、CD80、CD86和HLA-DR的表达及CIK和DC-CIK中CD3+CD56+细胞比率均较肿瘤患者来源的高;CIK和DC-CIK杀伤作用对A549细胞和肺腺癌原代细胞均具有杀伤活性,且随效靶比的升高而增强;DC-CIK杀伤活性较同来源的CIK强;同条件下,健康人来源的CIK或DC-CIK的杀伤活性较肿瘤患者来源强。[结论]健康人来源的CIK和DC-CIK对肺癌细胞杀伤活性优于肿瘤患者自身来源的CIK和DC-CIK。
