摘要
①目的探讨结缔组织生长因子在慢性心力衰竭大鼠心肌的表达及慢性心力衰竭心室重构和心肌纤维化可能的发病机制。②方法用腹主动脉结扎的方法进行慢性心力衰竭模型的制作。将实验大鼠随机分为4组:假手术组,心衰1天组,心衰7天组,心衰14天组。用免疫组化的方法对结缔组织生长因子在4组的左室心肌的分布及表达进行半定量分析。用Masson染色法观察左室心肌的胶原沉积情况。③结果在心力衰竭的不同阶段,大鼠的心室肌结缔组织生长因子表达明显增加(P<0.05);心衰后的各个阶段心肌的胶原沉积明显多于假手术组。④结论结缔组织生长因子参与了慢性心力衰竭心肌纤维化的过程。
Objective To investigate the expression of the connective tissue growth factor (CTGF) in the ventricular myocardium in chronic heart failure rats and the potential pathogemesis of the ventricles reconstitution and myocardial fibrosis in chronic heart failure. Methods Chronic heart failure rat models were established by abdominal aorta ligation. The experimental rats were divided into four groups: sham -operated groups and heart failure 1 day, 7 days and 14 days. CTGF were evaluated by immunohistochemical staining in the first, seventh and fouteen days after operation to explore their distributions and expressions in the left ventricular interstitial tissue. The Masson stainin was used for determining total collagen in the left ventriclar interstial tissue. Results The expression of CTGF in the ventaricular were significantly increased at different stages of the heart failure( P 〈0.05). Compared with sham - operated rats, total collagen at different stages of the heart failure were significantly increased. Conclusion The connective tissue growth factor might play an important role in the development of myocardial fibrosis in heart failure.
