摘要
目的观察葡萄籽提取物(GSE)对免疫性肝损伤的保护作用。方法分别制备卡介苗(BCG)联合脂多糖(LPS)诱导的大鼠原代肝细胞免疫损伤模型和小鼠免疫性肝损伤模型,测定细胞培养上清和血清中的天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)活力及一氧化氮(NO)水平,肝组织超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量,并进行肝脏病理组织学检查。结果在体外实验中,正常对照组大鼠肝细胞培养上清AST,ALT活力和NO水平24 h内未见明显变化,BCG+LPS损伤组大鼠肝细胞培养上清AST,ALT活力和NO水平在6、12和24 h较正常大鼠明显升高,加入LPS的同时分别加入GSE 5,20和80 mg/L均可不同程度地使升高的AST,ALT活力和NO水平下降。GSE上述作用与阳性对照药肝立欣(Grz)相似。体内实验表明,GSE 50,250和500 mg/kg(ig)均可明显降低免疫性肝损伤模型小鼠血清AST,ALT活力和肝组织MDA含量,升高肝组织SOD活力,并不同程度的减轻小鼠肝脏损伤。结论GSE对免疫性肝损伤具有明显的保护作用。
Objective To study the protective effect of the extract isolated form grape seed (GSE) on immunological liver injury. Methods Bacillus Cahnette Gnerin (BCG) plus lipopolysaceharides (LPS) were used to induce immunological primary hepatocyte injury model of rats in vitro and liver injury model of mice in vivo. The glutamate oxaloacetate transaminase (AST)and glutamate pyruvate transaminase (ALT) activities, and nitric oxide (NO)contents in cultured supernatant of cells and serum were mearsured, The superoxide dismutase(SOD)activity, malondialdehyde(MDA)contents, and histopathological changes in mouse liver tissue were also detected. Results In vitro, AST and ALT activities and NO contents in the hepatoeyte supematant of normal rats had no significant change after cultured for 24 h. gqlereas AST and ALT activities and NO contents in the supernatant of BCG sensitized (iv on tail, 8 mg per rat) rat hepatocytes cultured with LPS (10 mg/L) were increased after incubated for 6, 12 and 24 h, respectively. GSE (5, 20,80 rag/L) could reverse these changes when added with H^S on the same time, which was similar with glycyrrhizinate. In vivo, GSE (50, 250, 500 mg/kg)significantly inhibited the increase of the activities of AST and ALT in serum and MDA levels in liver tissue of mice induced by BCG (iv on tail, 1 mg per mouse) plus LPS (iv on tail, 7.5 btg per mouse). Otherwise, GSE improved the decrease of liver SOD activities and histopathological changes. Conclusion GSE has protective effect on immunological damage of liver.
