摘要
目的:筛选松茸多糖的最佳提取条件。方法:用水提取松茸多糖,以浸提温度、浸提时间、料水比和浸提次数为主要因素,采用L9(3^4)正交试验进行提取工艺的优化;用苯酚一硫酸法测定多糖含量后计算多糖得率。结果:影响松茸多糖提取的主要因素由大到小依次为浸提时间〉浸提次数〉浸提温度〉料水比。结论:通过正交试验及其验证试验确定的松茸多搪的最佳提取条件为:浸提时间3h,浸提温度90℃,浸提次数2次,料水比1:30,在此条件下,松茸多糖得率可达12.24%。
Objective: To screen the optimum conditions for extracting polysaccharide of T. matsutakes. Method: The polysaccharide of T. matsutakes was extracted with distilled water. The extraction temperature, extraction time, extraction ratio, extraction times were used as main factors, the L9 (3^4) orthogonal test was used for optimizing the extraction technology. The yield rate of polysaccharide was calculated after the polysaccharide content was detected by phenol-vitriol. Result: The main factors of effecting on the extraction for the polysaccharide of T. matsutakes from big to small were extraction time 〉 extraction times 〉 extraction temperature 〉 extraction ratio. Conclusion: The best conditions for extracting the polysaccharide by the orthogonal test and verification test was as follows: the extraction time for 3h,the extraction times for 2,the extraction temperature for 90℃, the extraction ratio for 1:30, and the yield of polysaccharide from T. matsutakes can achieve 12.24%.
