摘要
通过紫外诱变、硫酸二乙酯诱变及复合诱变,对南极假丝酵母菌种进行了选育,筛选出一株高产菌株UDAan-1,其酶活达到42.7 U.mL-1,为原始菌株的1.8倍。在摇瓶条件下对发酵培养温度、初始pH值等南极假丝酵母诱变菌株产酶工艺参数进行了优化。最终确定了各发酵工艺参数:培养温度26℃,初始pH值6.5,发酵用菌种培养20h,接种量6%(体积分数),摇床转速200 r.min-1,250 mL摇瓶装液量50 mL,表面活性剂选用0.1%吐温-80(体积分数)。
The strain of Candida Antarctica was selected by ultraviolet mutation, diethyl sulfate mutation and combination mutation. A lipase-high-producing strain UDAan-1 was selected and its enzyme activity was 42.7 U · mL^-1 , which was 1.8 times of initial strain. The lipase producing craft parameter for Candida Antarctica mutation strain was optimized in shake flask. The craft parameter included fermentation temperature, incipient pH and so on. The fermentation craft parameter was determined at last. The fermentation temperature was 26 ℃ ,incipient pH was 6.5, the fermentation strain age was 20 h, inoculum size was 6% ,the rotation speed of rocking bed was 200 r · min^-1 ,50 mL fermentation broth was filled in 250 mL shake flask, and the elected surface active agent was 0.1% Tween-80.
