结核分枝杆菌耐砒嗪酰胺pncA基因突变的检测 被引量:1

摘要 目的了解青岛市结核分枝杆菌(MTB)耐砒嗪酰胺(PZA)分离株pncA基因的突变情况,探讨其与耐PZA之间的关系。方法应用BacT/ALERT 3D培养系统对25例MTB临床分离株进行常规培养和药敏检测,通过PCR扩增pncA全长基因,DNA序列分析检测pncA基因的突变情况。结果25株MTB临床分离株有22株扩增出含pncA全基因的序列,进行DNA序列分析表明有6例发生了pncA基因的突变,突变发生在pncA基因29、349、428、533、539、541位的核苷酸,并导致PZA酶氨基酸序列的改变。结论pncA基因突变是青岛市MTB临床分离株耐PZA的分子机制之一。 Objective To investigate the mutation of pncA gene in clinical isolates of Mycobacterium tuberculosis (MTB) and the relationship between pncA gene and pyrazinamide (PZA) resistance strains in Qingdao area. Methods A routine MTB culture and drug susceptibility were done by BacT/ALERT 3D system in 25 clinical isolates of MTB, the pncA gene was amplified by polymerase chain reaction (PCR) and its mutation was analyzed by DNA sequencing. Results Of the 25 isolates, 22 strains were positive pncA gene by PCR amplification, and the DNA sequence showed that six mutations occurred in 29,349,428,533, 539,541 codon of pncA gene, respectively, which resulted in changes of Pzase Amino acid sequence. Conclusion Mutation of pncA gene might be one of the major molecular mechanisms in PZA resistance of clinical isolates of MTB.
