摘要
目的观察高糖状态下,人类肾小球系膜细胞(GMC)增殖,细胞内游离钙,纤维连接蛋白(Fn)及白介素-6(IL-6)的变化。方法采用体外GMC培养,3H-胸腺嘧啶(3H-TdR)掺入和细胞计数,Fura-2双波长荧光法和ELISA法。结果高糖对GMC增殖有抑制作用,对Fn及IL-6的生成有促进作用,而对细胞内游离钙水平无显著影响。
Objective To observe the effect of high glucose on the proliferation of cultured human glomerular mesangial cells,the level of cytosilic free calcium[Ca^2+]i,the fibronectin(Fn) and interleukin6(IL-6)levels in the tissue cultured supernatantMethods The culture of human glomerular mesangial cells was established and the cellular proliferation was assessed using 3 Hthymidine incorporation and cell counting,production of Fn and IL6 was analysed by ELISA,and the [Ca^(2+)]i level was determined by Fura2/AMResults High glucose depressed 3 HTdR incorporation while increased the production of Fn and IL6,but had no significant effect on [Ca^(2+)]i levelConclusion Hyperglycemia may be anentiologic factor of diabetic nephropathy
