摘要
目的:建立离子色谱法测定谷胱甘肽等含酰胺基药物中游离铵盐的含量。方法:采用IonPac CS12A色谱柱(250mm×4mm,8μm),IonPac CG12A预柱(50mm×4mm,8μm),以18mmol·L-1甲烷磺酸溶液为淋洗液,流速1.0mL·min-1,电导检测器检测。结果:NH4+在0.203～4.07μg·mL-1浓度范围内有良好的线性关系,r=0.9995;最低检测限为0.025ng;平均加样回收率在92.1%～100.6%之间。结论:本法简便、快速、准确,灵敏度高,重复性好,适用于含酰胺基药物中游离铵盐的质量控制。
Objective：To establish an ion chromatography（IC） method for determination of free ammonium salt in glutathione and other drugs containing amide. Methods：A column of IonPac CS12A （250 mm · 4 mm,8 μm） and a guard column of IonPac CG12A （50 mm ×4 mm,8 μm） ,a eluent consisting of 18 mmol · L^-1 methane - sulfonic with a flow rate of 1.0 mL · min^-1, and a conductivity detector were adopted. Results： The linear range of the determination was 0.203 -4.07 μg · mL^-1 ,r =0.9995 ;The limit detection was 0.025 ng;The average recovery was from 92.1% to 100. 6%. Conclusion： The method is simple, rapid, accurate, reproducible and high sensitivity. It can be used for quality control of drugs containing amide group.
