摘要 目的初步研究microRNA-21(miR-21)在大鼠胚胎植入过程中的作用。方法利用Northern印迹和原位杂交分析了miR-21在大鼠胚胎植入前期和植入期的表达差异,检测miR-21在假孕、人工诱导蜕膜化和延迟着床激活大鼠子宫中的表达模式,分析了17β-雌二醇和孕酮对于大鼠子宫miR-21表达的影响。结果 miR-21在植入前期和植入期的大鼠子宫中存在差异表达,在大鼠妊娠第5天miR-21的表达量开始增加,其主要分布在植入位点内腔基质细胞,在第5.5天子宫miR-21的表达量达到最高。在假孕的D4~D7大鼠子宫中miR-21表达无差异,在延迟着床激活模型中大鼠子宫miR-21的表达量明显增加,miR-21在诱导蜕膜化的大鼠子宫中表达也显著增加,17β-雌二醇和孕酮可以抑制大鼠子宫miR-21的表达。这些结果提示在植入期miR-21的表达增加主要是由胚泡引起的,并且其参与了蜕膜化过程。结论在大鼠妊娠早期,miR-21的表达增加可能有利于胚胎植入的发生。 Objective: To study the expression and regulation of miR-21 in the process of embryo implantation of rats. Methods: The differential expressions of miR-21 during the pre-implantation and implantation period and the expression profile in uterus of pseudopregnancy, artificial decidualization, delayed-implantation rats were detected by Northern blot and in situ hybridization. The effects of estradiol-17β or progesterone on miR-21 expression in uterus of ovariectomized rats were also investigated. Results: MiR-21 was highly expressed in the subluminal stromal cells at implantation sites on day 5 of pregnancy. The miR-21 expression was not significantly changed in uterus of pseudopregnancy rat and significantly increased in the uterus of artificial decidualization and delayed-implantation rat. The miR-21 expression was inhibited by treatment of estradiol-17β or progesterone. Collectively, the results suggested that increased miR-21 expression was mainly induced by blastocyst and may involve decidualization during the window of implantation, which may be beneficial to embryo implantation in the early of pregnancy in rats. Conclusion: MiR-21 may play an important role during embroy implantation in the rats.
