骨巨细胞瘤基质金属蛋白酶-9表达及与肿瘤血管关系 被引量:2

Expression of matrix metalloproteinase-9 in giant cell tumor of bone and its relationship with tumor angiogenesis

摘要 目的:探讨骨巨细胞瘤基质金属蛋白酶-9(MMP-9)表达强度、临床病理特征及与CD34标染肿瘤内微血管密度(mi-crovessal density,MVD)的相关性。方法:免疫组化SP法检测43例骨巨细胞瘤中MMP-9表达强度及CD34标染的新生血管密度,分析MMP-9表达强度、临床病理特征与MVD的关系。结果:MMP-9表达强度在骨巨细胞瘤病理分级及临床分级中呈逐渐升高的趋势,组间比较差异有显著性。复发组MMP-9表达强度高于初发组(P<0.01),在肿瘤转移组MMP-9表达强度高于未转移组(P<0.01)。Spear-man相关分析提示MMP-9表达强度与MVD值呈正相关,相关系数为0.839。结论:MMP-9可能通过促进骨巨细胞瘤内新生血管的形成而影响肿瘤预后,MMP-9可作为预测骨巨细胞瘤发生发展的检测指标之一。 Objective:To observe the expression of matrix metalloproteinase-9 (MMP-9) in giant cell tumor of bone (GCT) and their relations with microvessel density stained by CD34. Methods: Specimens from 43 cases of GCT were included in the study. Expression of MMP-9 and CD34 of GCT were studied by immunohistoehemistry. Microvessel density (MVD) was counted by endothelial cells immunostained by anti-CD34 antibody. The clinicopathological characteristics of MMP-9 expression and its relationship with the MVD value were analyzed. Results:MMP-9 expression was gradually increased by the tumor histologieal grade and clinieal classification,and the intergroup comparisons were significant. MMP-9 expression in the recurrence group was higher than that in the primary group(P〈0.01). MMP- 9 expression in the metastasis group was higher than that in the none metastasis group(P〈0.01). Spearman correlation analysis showed that MMP-9 expression and MVD values were positively correlated with the correlation coefficient 0.839. Conclusion:MMP-9 may affect tumor prognosis through its neovascularization promotion of giant cell tumor of bone. MMP-9 can be used as an indicator of development of giant cell tumor of bone.
