摘要
萤火虫荧光素酶只在萤火虫体内产生,一般通过养殖和用化学法提取。研究了以基因工程菌Escherichiacoli-Lus-J-1为材料,从它的对数生长期发酵液中提取萤火虫荧光素酶。经检测其分子量为5×104,动力学性质研究表明它是非典型的Michaelis-Menten氏酶,对荧光素作用动力学曲线为S型,最适pH为7.5,最适温度为15℃,且对光和盐浓度非常敏感。
The luciferase can be produced in body and can be obtained by breeding firefly and chemical extract. The luciferase has been purified from fermentative liquid of Escherichia coilLusJ, genetic recombination strain. Its molecular weight is 50000. Dynamic studies demonstrated that it does not behave the character of typical MechaelisMenten enzyme. The dynamics curve of the luciferase for luciferin shows a S shape. Its optimal pH and temperature are 7.5 and 15℃, respectively. It is sensitive to light and salt concentration.
