摘要
目的:愈来愈多的证据显示Caspase家族蛋白酶在细胞凋亡执行程序中发挥重要功能。辐射可诱发肿瘤细胞凋亡,分离、鉴定辐射诱发肿瘤细胞凋亡中Caspase家族成员,是肿瘤细胞凋亡调控机制关键所在。方法:根据已知的Caspase家族蛋白保守的氨基酸序列,设计简并引物,逆转录-聚合酶链反应(RT-PCR),分离、鉴定参与辐射诱发细胞凋亡基因。结果:Caspase家族蛋白酶特异的抑制剂能阻抑射线诱发的细胞凋亡,并从照后6hU937细胞分离出caspase-3基因。结论:Caspase参与辐射诱发的细胞凋亡,caspase-3基因是射线诱发细胞凋亡的枢纽元件。
Objective: Increasing evidences suggest that caspase family proteases play an important role in the effector mechanism of apoptotic cell death. Radiation(IR) can induce apoptosis in tumor cells, so it is very important to isolate and identify the member of the caspase family proteases involved in IRinduced apoptosis, and this would contribute to the understanding of the mechanism responsible for apoptosis execution. Methods: A PCR approach to isolate genes for IRinduced apoptosis was developed. The approach used degenerated oligonucleotides encoding the highly conserved peptides that were present in all known caspases. Results: Protease inhibitors special for caspases could block the apoptotic cell death caused by IR, and caspase3 was isolated from irradiated human leukemia U937 cells. Conclusion: Caspases involve in IRinduced apoptosis, and caspase3 is the pivotal element of IRinduced apoptosis.
