摘要
目的观察姜黄素对磨损颗粒刺激炎性介质产生的影响,进一步探讨炎性介质表达调节机制。方法构建小鼠air-pouch动物模型成功后,于囊腔中注射高分子聚乙烯颗粒悬液。随机分为实验组和对照组,分别每日每次0.6ml,浓度为3.2g.L-1姜黄素溶液灌胃和等量6%的聚乙二醇溶液灌胃,共2wk,给药后2wk处死取材,进行组织学观察,采用ELISA法及半定量RT-PCR检测假体周围组织中TNF-浕、IL-1β、EMMPRIN、MMP-9的表达。Western blot检测囊壁组织核蛋白中NF-κBP65蛋白的表达。结果组织学观察发现对照组的囊腔范围及囊壁厚度明显大于给药组。HE染色囊壁中有大量炎性细胞,两组间炎性细胞计数差异有统计学意义;两组囊壁组织中TNF-浕、IL-1β、EMMPRIN、MMP-9、NF-κBP65表达有统计学意义。结论姜黄素可以抑制air-pouch动物模型界膜组织中TNF-浕、IL-1β、EMMPRIN、MMP-9的表达,可能是通过下调囊壁组织中核蛋白NF-κB完成的。
Aim To observe and investigate the mechanism of curcumin on the experssion of mediators of inflammation. Methods Using a murine air pouch model of inflammation, 24 Kunming mice were divided into 2 groups (12 per group) in the present study. The air pouches were injected with 3ml of suspension containing 1 ×10^8· ml^-1 UHMWPE particles. In the control group, the 0. 6 ml saline was perfused into the empty stomach daily for 2 weeks. 0. 6 ml Curcumin solution (3.2 g·L^-1) was perfused into the empty stomach daily for 2 weeks in the experimental group. At the end of 2 weeks the rats were sacrificed and all air-pouch was harvested , then histology and histomorphometry were carried out. TNF-α,IL-1 β,EMMPRIN and MMP- 9 were respectively tested by ELISA and semi-quantified RT-PCR,and the nuclear protein of NF-κBP65 was tested by Western blot. Results The histological analysis showed the air-pouch positioned subcutaneous at the back, and the air-pouch of control group was bigger than that of the experimental group. The number of inflammatory cells showed significant difference between control group and experiment group in statistics. The difference was significant in statistics when TNF-α, IL- 1β,EMMPRIN and MMP-9 in the air-pouch in the two groups were tested respectively by ELISA and semiquantified RT-PCR, the statistics of NF-κBP65 also showed significant difference between the two group. Conclusions Curcumin inhibits TNF-α, IL-1β, EMM- PRIN and MMP-9 expression in air-pouch model. Via reducing the expression of NF-κB, curcumin may becomes the medicine which prevents the osteolysis around the prosthesis.
