摘要
目的研究苦马豆素(α甘露糖苷酶Ⅱ抑制剂-swainsonine,SW)对宿主的免疫调理作用及其对胃癌生长及转移的抑制作用。方法给胃癌裸鼠原位种植模型饲养含SW3μg/ml饮水,每间隔2天换饲普通饮水2天,8周后处死实验动物,测量肿瘤大小,观察转移情况,测定脾细胞总数、脾细胞及外周血淋巴细胞NK活性。结果SW组肿瘤体积为0.42±0.26cm3,明显小于对照组1.60±0.36cm3,抑瘤率达73.8%(P<0.01);SW组肝及腹膜转移率(20%、30%)均明显低于对照组(80%、90%),抑制率分别达到75.0%和66.7%(P<0.01)。饲SW饮水后,裸鼠脾细胞总数明显增加,外周血淋巴细胞NK活性明显升高,脾细胞NK活性亦明显升高。结论SW有抑制人胃癌在裸鼠体内生长和转移的作用。
Objective To study the immunomodulating effect of Swainsonine (SW), an inhibitor of α mannosidase II, and the inhibiting effect on the growth and metastasis of gastric cancer. Methods Nude mice with human gastric cancer xenograft orthotopically implanted into the gastric wall were fed with drinking water containing 3 mg/L SW for two days alternating with that containing no SW for two days until sacrificed eight weeks after tumor implantation. Results The mean tumor volume in the SW treated group was significantly smaller than that of the control group (0.42±0.26 cm 3 vs 1.60±0.36 cm 3). Calculated by volume, the tumor growth was inhibited by 73.8%. Metastasis to the liver was observed in 20% and 80% of the SW treated and control mice, while metastasis to the peritoneum was seen in 30% and 90% of SW treated and control mice, respectively. The total number of splenic cells, and the NK cell activity both of PBL and splenic cells were all elevated. Conclusion SW is effective in inhibiting tumor growth and metastasis of human gastric cancer in nude mice. Enhanced immune response, among other things, may be involved in the anti tumor effect.
