摘要
从脂肪嗜热芽孢杆菌FH58和S183中分别分离到Ⅱ型的限制性内切酶BstFⅠ和BstSⅠ。经鉴定,BstFⅠ的识别顺序和酶切位点为A/AGCTT,与Hind Ⅲ一致;BstSⅠ的识别顺序和酶切位点为C/PyCGPuG,与AvaⅠ一致。经肝素琼脂糖凝胶提纯,每克湿菌体可分离到10000单位的BstFⅠ或24000单位的BstSⅠ。BstFⅠ在45℃中保温6h,比活力不变,在50℃中保温1h比活力下降50%;而BstSⅠ在55℃中保温6h,比活力不变,在70℃中保温1h,比活力仅下降10%。
Two type II restriction endonucleases, BstF I and BstS I, have been isolatedfrom Bacillus stearothermophilus FH58 and Bacillus stearothermophilus S183. Therecognition sequence and cleavage site of BstF I is A/AGCTT; and C/PyCGPuGin BstS I. Therefore. BstF I is the isoschizomer of Hind III and BstS I is the isos-chizomer of Ava I. These two enzymes can be easily purified with heparin-agarose. 10 000 units of BstF I and 24 000 units of BstS I can be purified per gramwet cell of FH58 or S183, respectively. They have different thermostability. BstFI and BatS I are stable under incubation at 45℃ for as long as 6 hours. After 1 hincubation at 50℃ the relative activity of BstF I was reduced by 50%, whereas therelative activity of BstS I was reduced only by 10% after 1 h incubation at 70℃.
