摘要
目的对3种常规血浆总二氧化碳(TCO2)测定方法进行评估,观察重量法配制的水质NaHCO3标准液的互通性。方法通过观察电极法、以还原性辅酶Ⅰ变体(Thio-NADH)为底物的酶促动力学法和以NADH为底物的酶促终点法的线性范围、精密度、回收率、抗干扰能力和偏倚对3种方法的分析性能进行评估。通过对两两方法间40份新鲜血浆标本TCO2的测定结果作相关分析,观察水质NaHCO3标准液测定值是否位于95%的可信区间,判别自配标准液的互通性。结果3种方法批内不精密度(n=20)〈3.3%,批间不精密度(n=20)〈5.2%。电极法和以Thio—NADH为底物的酶法试剂线性范围为5—50mmol/L;以NADH为底物的酶法试剂线性范围为5—45mmol/L。3种方法的回收率分别为95%~106%、99%-103%和95%-106%。以电极法作为对照,以Thio—NADH为底物的酶法试剂测定的相对偏倚在-6.45%~2.51%之间;以NADH为底物的酶法试剂测定的相对偏倚在-6.34%~6.36%之间。TBil〈300μmol/L、Hb〈10g/L、TG〈25mmol/L对电极法和以Thio-NADH为底物的酶法试剂测定血浆TCO2结果无影响;TBil〈300μmol/L、Hb〈5g/L、TG〈13mmol/L对以NADH为底物的酶法试剂测定血浆TCO2结果无影响。3种方法测定NaHCO3水质标准液浓度测定值落在95%的可信区间内。结论3种方法具有良好的分析性能,检测结果等效一致;重量法配制的NaHCO3水质标准液具有互通性。
Objective To evaluate the analytical performance of three conventional methods for the determination of total plasma carbon dioxide (TCO2 ) and observe the eommutability of sodium bicarbonate ( NaHCO3 ) standard solution prepared by weighting method. Methods Three methods:electrode method, kinetics enzymatic method using NADH variant (thio-NADH) as substrate and endpoint enzymatic method using NADH as substrate, were evaluated on linearity, precision, recovery rate,interference and bias. The TCO2 in 40 fresh plasma specimens were determined by the three methods respectively, and the correlation of various methods were analyzed. 95% confidence interval was set to observe the eommutability of self-prepared standard solution. Results The within-run imprecision was 3. 3% (n = 20 ), and between-run imprecision was 〈 5.2% (n = 20 ) for three methods. The lineariries of electrode method and enzymatic method using NADH variant ( thio-NADH ) as substrate were similar, from 5 mmol/L to 50 mmol/L. The linear range of enzymatic method using NADH as substrate was from 5 mmol/L to 45 mmol/L. The recovery rates of the three methods were from 95% to 106%, 99% to !03%, and 95% to 106% ,respectively. When the electrode method was used as the control method, the relative bias of enzymarie method using NADH variant as substrate was from - 6.45 to 2.51%, and the relative bias of enzymatic method using NADH as substrate was from -6.34% to 6.36%. The conditions: bilirubin 〈 300μmol/L, haemoglobin 〈 10 g/L and triglyeeride 〈 25 retool/ L had no interference on the determining of plasma TCO2 by electrode method and enzymatic method using NADH variant as substrate. The conditions:haemoglobin 〉 5 g/L and triglyceride 〉 13 mmol/L had negative interference by enzymatic method using NADH as substrate. The concentrations of the self-prepared NaHCO3 standard solurion determined by the three methods were all within the 95% confidence interval. Conclusions The three evaluated methods have good performance and consistency in determining the total plasma CO2, and the NaHCO3 standard solution prepared by weighting method exhibits eommutability.
