BMI-1在不同时期表皮鳞癌细胞中的表达

Expression and significance of BMI-1 in proliferation of Squamous Cell Carcinoma

摘要 目的研究全反式维甲酸(ATRA)对表皮鳞癌细胞株A431细胞凋亡的影响,并观察干细胞更新因子BMI-1在鳞癌A431细胞系不同分化时期的表达变化,以明确BMI-1在皮肤鳞癌细胞增殖分化中的功能。方法体外培养A431细胞,分别于ATRA作用即刻、24 h及48 h收集细胞,流式细胞仪检测不同状态下细胞凋亡变化,west-ern blot法检测不同时期BMI-1蛋白表达差异。结果随ATRA作用时间的延长可以显著提高A431细胞的凋亡率。同时24 h及48 h BMI-1表达逐渐减少,与即刻组之间差异有统计学意义(P<0.05)。结论ATRA可抑制鳞癌细胞A431增殖,并影响BMI-1在肿瘤细胞不同增殖状态的表达,同时说明干细胞更新因子BMI-1在维持皮肤鳞状细胞癌增殖中可能起关键作用。 Objective To observe effect of all -trans -retinoic acid (ATRA) on squamous cell carcinoma(SCC) cell line A431 and expression of BMI- 1, hematopoietic stem cell renewal factor, in the A431 treated by ATRA, Futher, role of BMI - 1 in proliferation of SCC will be evaluated. Methods After exposed to ATRA for Oh ,24h and 48h, A431 were harvested to detect the The cell apoptosis through flow cytometry and expression of BMI - 1 by western blot. Results The apoptosis rate was increased after ATRA treatment for 24h, 48h, along with BMI - 1 expresssion decreased obviously ( P 〈 0.05 ). Conclusion ATRA could suppress the proliferation of A431 and expression of BMI - 1. Moreover; we proposed that stem cell renewal factor BMI -1 plays a key role in proliferation of SCC.
