急性白血病患者多药耐药基因表达的检测及意义 被引量:1

The detection and significance of multidrug resistance (mdr_1) gene expression in acute leukemia

摘要 为研究急性白血病患者多药耐药基因(MDR1)表达与临床耐药、预后判断等关系,应用逆转录酶/多聚酶链反应(RT-PCR)检测了36例急性白血病患者及10例正常人骨髓细胞MDR1基因的表达,并以MDR/βMG≥0.2定为MDR1mRNA阳性,结果显示,10例正常人MDR1mRN表达均阴性。初治患者中MDR1mRNA阳性率为25%(4/16),而复发和未缓解患者则达80%(16/20)。化疗缓解率在MDR1mRNA阴性及阳性的急性白血病患者中分别为94%和20%,提示MDRmRNA表达与化疗反应显著相关,MDR1mRNA阳性化疗缓解率低,预后差,可将MDR1mRNA阳性作为化疗耐药指标。本研究为逆转克服多药耐药性药物的应用及个体化治疗方案的选择提供了一定的依据。 By using reverse retrotranseriptade polymerase cha'n reactio (RT-PCR)teehnique,the expression of MDR1 gene in 36 patients with acute leukemia was asayed.The results showed that there are higher levels of MDR1mRNA in 80%(16/20)of the relapsed andrefractory patiennts and only in 25%(4/16)of untreated patients.The chemotherapy effective rate was 94%(15/16)in the MDR1gene expression negative patients,and 20%(4/20)of the positive patients There was correlationship between MDR1 gene expression and cheotherapeutic effect.It suggested that the highet level of MDR1mRNA leads to drug resistance and mighe beapredictive index for poor prognosis in acute leukemia patients.
