摘要
以广西地不容叶片、茎段为材料,采用正交设计法研究6-BA、2,4-D、NAA 3种植物生长调节剂组合对广西地不容愈伤组织诱导的影响。结果表明:不同组合均能诱导广西地不容叶片和茎段产生愈伤组织,但诱导效果存在一定差异;除6-BA、NAA对茎段愈伤组织相对生长速率影响显著外,3种生长调节剂对叶片、茎段愈伤组织诱导率及叶片愈伤组织相对生长速率影响均不显著。诱导叶片、茎段愈伤组织形成的适宜培养基分别为6-BA 0.5mg/L+2,4-D0.4mg/L+NAA 0.2mg/L和6-BA 1.0mg/L+2,4-D 0.8mg/L+NAA 0.8mg/L,有利于提高愈伤组织生长速率的适宜培养基是:叶片6-BA 0.5mg/L+2,4-D 0.4mg/L+NAA 0.2mg/L,茎段6-BA 2.0mg/L+2,4-D 0.2mg/L+NAA 0.4mg/L。研究结果为广西地不容再生体系的建立奠定基础。
In the experiment, leaves and stems were used as explants to study the effects of plant growth regulator combinations including 6-BA, 2,4-D and NAA on the callus induction of Stepbania kwangsiensis Lo. by orthogonal design. The results showed that all the combinations could induce callus from leaves and stems, but the induction effects were different. Adding 6-BA and NAA to the medium had significant effects on the relative growth rate of callus derived from stem, while all regulators had no significant effects on the induction rate of callus derived from leaves and stems and the relative growth rate of callus derived from leaves. To leaf and stem, the optimal medium of callus induction was 6-BA 0.5mg/L+2,4-D 0.4mg/L+NAA 0.2mg/L and 6-BA 1.0mg/L+2,4-D 0.8mg/L+NAA 0.8mg/L, respectively. The optimal medium which was beneficial to increase the growth rate of callus derived from leaf and stem was 6-BA 0.5mg/L+2,4-D 0.4mg/L+NAA 0.2rag/L, and 6-BA 2.0mg/L+2,4-D 0.2mg/L+NAA 0.4mg/L These results provided the basis on establishing the regeneration system of Stephania kwangsiensis Lo.
