聚β-羟基丁酸酯材料的鼠伤寒沙门氏菌回复突变试验 被引量:7

摘要 背景:鼠伤寒沙门氏菌回复突变试验具有一定的预测致癌物的能力,其敏感性、特异性、准确性较高。目的:通过对聚β-羟基丁酸酯材料进行鼠伤寒沙门氏菌回复突变试验,评价该材料的潜在致突变性。设计、时间及地点:体外对比观察实验,于2009-02/03在国家沈阳新药安全评价研究中心实验室完成。材料:聚β-羟基丁酸酯(10mm×10mm)由中国科学院金属研究所提供,组氨酸营养缺陷型鼠伤寒沙门氏菌TA97,TA98,TA100,TA102由沈阳市安全评价中心遗传毒性室提供。方法:采用标准平板掺入法,计数TA97,TA98,TA100,TA102标准测试菌株在4个不同浓度浸提下,计算于37℃培养48h后的回复突变菌落数。加S9混合液作为体外代谢活化系统。主要观察指标:各平皿细菌回复突变菌落数,凡高于阴性对照2倍以上者即为阳性结果。结果:聚β-羟基丁酸酯细菌回复突变试验显示,各剂量组细菌回复突变数小于阴性对照组细菌回复突变数的2倍。各菌株对应各剂量组无论代谢活化与否,皆为阴性试验结果。结论:聚β-羟基丁酸酯经细菌回复突变试验未见潜在致突变性。 BACKGROUND: Salmonella typhimurium Ames test has the ability to predicting carcinogen and high sensitivity, specificity, and accuracy. OBJECTIVE: To evaluate the potential mutagenicity of poly-beta-hydroxybutyrate (PHB) via salmonella typhimurium Ames test. DESIGN, TIME AND SETTING: An in vitro contrast study was performed at the laboratory of Shenyang Xinyao Safety Evaluation Center from February to March 2009. MATERIALS: PHB sized 10 mm × 10 mm was provided by Metal Institute of Chinese Academy of Science; histidine-auxotroph spontaneous revertants TA97, TA98, TA100, and TA102 were provided by Genetoxic Laboratory of Shenyang Safety Evaluation Center. METHODS: In the fiat-plate incorporation test of salmonella typhimurium, the average number of spontaneous revertants of TA97, TA98, TAI00, and TA102 at four concentrations was calculated after incubation at 37 ℃ for 48 hours. S9 mixed liquid was used as metabolism activation system in vitro. MAIN OUTCOME MEASURES: The number of spontaneous revertants which was twice more than negative controls was considered as the positive one. RESULTS: Salmonella typhimurium Ames test demonstrated that the number of spontaneous revertants was twice less than negative control group. The result of Ames test for PHB was negative. CONCLUSION: PHB has no potential mutagenicity via salmonella typhimurium Ames test.
