摘要
目的比较正常结肠粘膜与结肠癌间基因表达的差异。方法以人正常结肠粘膜和结肠癌为研究对象,建立信使RNA差异显示法筛选二者间差异表达的基因信息。结果建立了稳定的差异显示技术,初步应用于结肠癌的研究,获得一个在结肠癌高表达而正常组织无表达的基因片段。结论信使RNA差异显示法是研究结肠癌分子生物学基础的有力工具,所获基因片段可能是与结肠癌相关的癌基因。
Objective To compare the difference of gene expression between human normal colon mucosa and colon cancer. Methods mRNAs from human normal colon mucosa and colon cancer were compared by means of the technique of mRNAs differential display. Results The technique of mRNAs differential dis-play was established, and applied in clinical practice. A cDNA fragment obtained had a high expression in colon cancer, but had no expression in normal colon mucosa. Conclusion The technique of mRNAs dif- ferential display is a helpful tool in the molecular biological study of colon cancer. The cDNA fragment might be a colon cancer related oncogene.
