摘要
分别以东北刺人参叶芽和带有叶柄的幼叶为外植体,研究6-BA浓度对东北刺人参体细胞胚状体诱导的影响,并利用扫描电镜对体细胞胚胎发生的形态学进行了观察。结果表明:以带有叶柄的东北刺人参幼叶为外植体,在MS培养基上附加4.0 mg/L 6-BA和0.5 mg/L NAA,可获得健状的胚状体,并且胚状体经过球形胚、心形胚、鱼雷形胚和子叶形胚的胚胎发育时期,最终形成可发育成完整小植株的成熟胚。
The leaves with leafstalk and buds of Oplopanax elatus Nakai were taken as explants and cul- tured in vitro. Large and healthy embryoids were obtained on MS medium supplement with 4.0 mg/L, 0.5 mg/L NAA from buds. The development of somatic embryos of Oplopanax elatus Nakai was observed by scanning electron microscopy. The results showed that the embryoids were developed from global, heart-shaped, torpedo-shaped and cotyledon embryo, then matured and formed planet.
