摘要
目的:观察水飞蓟宾对人肝星状细胞LX-2凋亡与α1(I)胶原mRNA表达的影响。方法:人肝星状细胞株LX-2分为对照组与实验组,对照组以体积分数10%胎牛血清处理,实验组分别以体积分数10%胎牛血清和终浓度分别为6.25mg/L、12.5mg/L、25mg/L、50mg/L的水飞蓟宾处理48h后,应用流式细胞仪Annexin/PI双染方法检测细胞凋亡,应用RT-PCR检测各组细胞α1(I)胶原mRNA的表达。结果:12.5mg/L、25mg/L、50mg/L水飞蓟宾可促进LX-2细胞凋亡,细胞凋亡率随水飞蓟宾浓度增加而升高(P<0.05);不同浓度的水飞蓟宾明显降低LX-2细胞α1(I)胶原mRNA的表达(P<0.05)。结论:水飞蓟宾可促进肝星状细胞凋亡,抑制α1(I)胶原mRNA的表达。
Aim : To investigate the effects of silibinin on apoptosis and α1 (Ⅰ) collagen mRNA expression in cultured human hepatic stellate cells (HSCs) LX-2 in vitro. Methods:HSCs were divided into control group and experiment group. HSCs in the control group were treated with 10% fetal calf serum (FCS)/DMEM and HSCs in the experiment group were treated with 10% FCS/DMEM and silibinin with different concentrations (6.25 mg/L, 12.5 mg/L,25 mg/L,50 mg/L) for 48 hours. Flow cytometry was used to evaluate the apoptosis of HSCs using Annexin/PI-staining and RT-PCR was used to determine the etl (1) collagen mRNA expression. Results: Silibinin with different concentrations( 12.5 - 50 rag/L) could promote HSCs apoptosis and the apoptosis rate raised with the concentration of silibinin ( P 〈 0.05 ). Silibinin could significantly decrease the α1 (Ⅰ)collagen mRNA expression (P 〈 0. 05 ). Conclusion: Silibinin can promote apoptosis and inhibit the α1 (Ⅰ) collagen mRNA expression of HSCs.
