摘要
目的探讨TGFα、EGF及其受体EGFR在小鼠小肠上皮受照射后再增殖中的作用。材料与方法雄性C3Hf/Kam小鼠5Gy全身一次性照射后,不同时间杀死小鼠取出空肠,常规HE染色高倍下计数隐窝细胞数和分裂相;并以免疫组化法定量测定隐窝区和绒毛区TGFα、EGF和EGFR的表达。结果照射后3~4天小鼠小肠上皮隐窝细胞及分裂相均达到高峰;而TGFα、EGF和EGFR在照射后第1天即在隐窝区达到高峰,随之下降,其表达变化时间上领先于细胞分裂。结论TGFα、EGF和EGFR的表达可能在小鼠小肠上皮受照射后的再增殖过程中起着重要的作用。
Purpose To study the role of expression of TGF-α,EGF and EGF-R in the repopulation of mouse jejunum following irradiation. Materials and Methods Using the jejunum of C3Hf/Kam mouse,the mitotic index and the number of crypt cells were measured at different times after single whole body irradiation of 5 Gy.At the same time,the expression of TGF-α,EGF and EGF-R was also tested quantitatively by automated immunohistochemical staining method. Results The mitotic index and the number of crypt cells reach peaks three-four days after irradiation.Meanwhile,the expression of TGF-α,EGF and EGF-R in crypt peak one day after irradiation. Conclusion The expression of TGF-α,EGF and EGF-R may play an important role in the repopulation of mouse jejunum following irradiation.
