多重评价液氮冷冻加被动强化运动建立股骨头坏死模型 被引量:1

Multiple Systems for Evaluatation of Models of Osteonecrosis of Femoral Head by Liquid Nitrogen Frozen Plus Passive Forced Motion

摘要 目的多重评价液氮冷冻加被动强化运动法建立股骨头坏死模型的效果。方法选用毕格犬8只,用液氮冷冻加术后被动强化运动法制备股骨头坏死模型(实验侧),对侧对照(对照侧)。术后1、4、8、16周取股骨头标本行苏木精-伊红染色;术后1、8、16周行MRI扫描;术前及处死前行数字减影血管造影术(DSA)。结果实验侧均出现不同程度股骨头坏死表现,且术后16周标本呈轻度塌陷;MRI示术后8周实验侧T2WI出现高信号,内部散在低信号影,术后16周低信号影进一步扩大;DSA示术后16周股骨头各支血管呈现不同的程度的狭窄或堵塞,并可见新生血管经股骨颈、髓腔等方向进入股骨头,但大多未能到达坏死区域。结论液氮冷冻加被动强化运动法建立的股骨头坏死模型符合股骨头坏死的病理变化,是一种比较理想的造模方法。 Objective To evaluate an animal model of osteonecrosis of femoral head. Methods Eight Beagle dogs were selected,and the model of osteonecrosis of femoral head was established by liquid nitrogen frozen plus passive forced motion.Meanwhile ,the left side served as control groups.Histological slices of femoral head were harvested on postoperative weeks 1,4,8 and 16 for HE staining.The osteonecro- sis of femoral head was observed by magnetic resonance imaging at 1,8 and 16 weeks after operation.DSA was performed before operation and sacrificing the animals to compare the pre-and post-operation blood supply surrounding femoral head. Results Pathological sections suggested that all operated femoral heads showed osteonecrosis at different levels ,moreover,slightly collapse was appeared in the 16-week specimen; MRI showed the scattered low signals were found in the operated femoral heads at the 8 week on T2WI, which were enlarged at the 16 week;DSA images displayed that stenosis or blockage at different levels of blood vessels which supply the femoral head,meanwhile,a plenty of new blood vessels grew into the femoral head through the neck of femur,and bone marrow,but most of them could not reach the necrotic area. Conclusion Liquid nitrogen frozen plus passive forced motion is an ideal method to establish the animal model of osteonecrosis of femoral head that consists with the pathological change of osteoneerosis of the femoral head.
