趋化因子CCL5促进乳腺癌细胞转移机制初探 被引量:3

摘要 目的:探讨趋化因子CCL5在人乳腺癌细胞MCF-7转移中的作用及其机制。方法:RT-PCR法测定MCF-7细胞中趋化因子CCL5的受体CCR5的表达;趋化小室检测不同浓度CCL5的作用下MCF-7细胞的侵袭活性;Western blot检测MCF-7细胞中Ezrin蛋白表达随CCL5趋化时间的变化。结果:人乳腺癌细胞MCF-7表达CCR5 mRNA,不同浓度(0.1-100μg/L)CCL5促进MCF-7细胞侵袭能力不同,Ezrin蛋白表达随CCL5促进乳腺癌细胞侵袭时间的延长明显增加。结论:趋化因子CCL5促进人乳腺癌细胞的趋化与侵袭,Ezrin蛋白在此过程中可能起着重要作用。 To study the chemotaxis and invasion of breast cancer MCF-7 cells induced by CC chemokine-5 (CCL5) and the mechanism of the process. Methods.. Reverse transcription-PCR analysis of CC chemokine receptor 5 (CCRS) was performed in the breast cancer cell line MCF-7. The invasion of MCF-7 cells was assayed by Borden chamber experiment. Western blot were performed to examine T-Ezrin and P-Ezrin expression in the MCF-7 cells responded to CCL5 at different times. Results: Breast cancer cell line MCF 7 expressed CCR5 mRNA. The cells responded chemotactically to CCL5 was dose-dependent. The expressions of phosphorylated and total Ezrin were significantly up-regulated in MCF 7 cells after induced by CCLS. Conclusion.. Chemotaxis and invasion of breast cancer cells can be facilitated by CCLS. Ezrin might play a potential role in the process of migration, invasion, and metastasis of breast cancer cells responding to the chemokine CCLS.
