摘要
目的:探讨多胺生物合成关键酶ODC的不可逆抑制剂DFMO对人早幼粒白血病细胞HL60生长及凋亡的影响。方法:细胞形态观察、流式细胞分析、DNA凝胶电泳及免疫细胞化学等。结果:DFMO不仅能阻止HL60细胞生长,而且能诱导HL60细胞凋亡,同时引起凋亡抑制基因bcl2表达下调。此外用外源性腐胺与DFMO同时处理HL60细胞可阻止DFMO所引起的上述变化。结论:DFMO阻断HL60细胞的多胺生物合成可在一定程度上使HL60细胞恶性表型发生逆转,提示多胺生物合成抑制是治疗肿瘤一种值得进一步研究的途径。
Objective:To observe the effect of DFMO,an inhibitor of ODCa key enzyme of polyamine biosynthesis,on the growth and apoptosis of HL60 cells.Methods:Cell morphological assay,flow cytomeric analysis、DNA fragmentation and immunocytochemical staining were used.Results:DFMO not only arrested HL60 cells growth,but also induced their apoptosis which was associated with down regulation of bcl2 gene expression.These changes resulted from DFMO were prevented,however,when the cells were treated by DFMO along with exogenous putrescine.Conclusion:The control of polyamine biosynthesis in HL60 by DFMO could reverse its malignant phenotypes to a certain extent and it is a worth search approach for tumor therapy.
