多胺生物合成抑制对HL-60细胞生长及凋亡的影响

摘要 目的:探讨多胺生物合成关键酶ODC的不可逆抑制剂DFMO对人早幼粒白血病细胞HL60生长及凋亡的影响。方法:细胞形态观察、流式细胞分析、DNA凝胶电泳及免疫细胞化学等。结果:DFMO不仅能阻止HL60细胞生长,而且能诱导HL60细胞凋亡,同时引起凋亡抑制基因bcl2表达下调。此外用外源性腐胺与DFMO同时处理HL60细胞可阻止DFMO所引起的上述变化。结论:DFMO阻断HL60细胞的多胺生物合成可在一定程度上使HL60细胞恶性表型发生逆转,提示多胺生物合成抑制是治疗肿瘤一种值得进一步研究的途径。 Objective:To observe the effect of DFMO,an inhibitor of ODCa key enzyme of polyamine biosynthesis,on the growth and apoptosis of HL60 cells.Methods:Cell morphological assay,flow cytomeric analysis、DNA fragmentation and immunocytochemical staining were used.Results:DFMO not only arrested HL60 cells growth,but also induced their apoptosis which was associated with down regulation of bcl2 gene expression.These changes resulted from DFMO were prevented,however,when the cells were treated by DFMO along with exogenous putrescine.Conclusion:The control of polyamine biosynthesis in HL60 by DFMO could reverse its malignant phenotypes to a certain extent and it is a worth search approach for tumor therapy.
