摘要
以盾叶薯蓣(Dioscorea zingiberensis C.H.Wright)无菌苗的茎段和叶片为外植体,研究了适于盾叶薯蓣再生的外植体类型和植物生长调节剂组合,建立了一个高频的离体再生体系.结果表明:以无菌苗茎段为外植体,在2.0—3.0mg·L^-1 6-苄氨基腺嘌呤(BA)作用下可诱导产生致密的胚性愈伤组织;转移到含2.0~3.0mg·L^-1 BA和0.2—0.5mg·L^-1 α-萘乙酸(NAA)的分化培养基上可迅速分化出大量的丛生芽,分化率达90%以上;无根苗在1/2MS+NAA 0.1~0.2mg·L^-1的生根培养基上生根率超过90%,移栽成活率达70%以上.
Taking the in vitro cultured leaves and stems from Dioscorea zingiberensis as explants, the effects of explants type and hormone combination in medium on shoot regeneration of Dioscorea zingiberensis were studied in the experiment. A high frequency regeneration system of Dioscorea zingiberensis using plantlets in vitro as explants was established. The results showed that the stems of sterile plantlets induced from single node segments could be induced compact embryonic calluses by 2.0 - 3.0 mg ·L^-1 BA, and more than 90% of embryonic calluses initiated adventitious buds within 30 d, then adventitious buds produced plants whose branches and leaves were flourish on the differentiation medium with BA 2.0 - 3.0 mg · L^- 1 and NAA 0.2 - 0.5 mg · L^-1 Splitting branches which transplanted onto the medium of 1/2 MS + NAA 0.1 -0.2 mg · L^-1 got a high rooting rated up to 90%, and transplanting survival rated up to 70%. The establishment of high frequency regeneration system of Dioscorea zingiberensis laid a oundation for genetic transformation research of Dioscorea zingiberensis.
