褪黑素在猪精液保存中的抗氧化作用 被引量:21

摘要 为探讨褪黑素(MLT)在猪精液保存中的抗精子氧化损伤效应,采用分步筛选的方法,首先在15℃保存稀释精液中分别添加1×10-6mol/L(M1)、1×10-5mol/L(M2)、1×10-4mol/L(M3)、1×10-3mol/L(M4)和1×10-2mol/L(M5)MLT,以不添加MLT为对照组(M0),检验并选择MLT有效浓度用于15℃保存的离心精液,检验并选择MLT最佳浓度用于冷冻精液。结果表明:在保存6 d的稀释精液中,M1、M2和M3组精子活率(TMS)、质膜完整性(PMI)和顶体完整性(NAR)均高于M0,M2最高,M4、M5则显著低于M0(P<0.05);丙二醛(MDA)浓度均低于M0,并以M2为拐点先降后升;选择M1、M2和M3用于离心精液保存,6 d后TMS和NAR均高于对照组,且M2与对照差异显著(P<0.05),MDA浓度则逐渐降低且与对照差异显著(P<0.05);选择M2用于精液冷冻保存,解冻精液TMS、PMI、NAR和线粒体膜电位(Mt-MP)显著提高(P<0.05),MDA浓度显著降低(P<0.05)。可见,添加适宜浓度MLT能够抑制稀释、离心及冷冻保存猪精液中的精子氧化损伤,显著提高精液保存质量。 The effect of anti-oxidative damage of melatonin (MLT) was evaluated on preserved boar semen. Firstly, MLT was added to diluted boar semen at 15 ℃ with five concentration of 1 × 10^- 6(M1 ) , 1 × 10^-5(M2) , 1 × 10 ^-4( M3 ), 1 ×10^- 3 (M4) and 1 × 10^ -2 mol/L ( M5), then the effective concentration of MLT were determined and applied to centrifugal boar semen, and the optimal concentration of MLT were applied to freezing media of semen cryopreservation. Results showed that the percentages of total motile sperm (TMS), plasma membrane integrity (PMI) and normal acrosomal ridge (NAR) in group M1, M2 and M3, especially in group M2, were higher than those of control, but significantly lower in group M4 and M5 than control (P 〈 0. 05); meanwhile, the concentration of MDA gradually decreased in group M1 and M2, then increased in group M3, M4 and M5, and it was lower than control in five groups at 3 d during the preservation. For the centrifugal boar semen, the percentages of TMS and NAR in group M1, M2 and M3 were higher than control,and there was a significant difference between group M2 and control ( P 〈 0. 05 ) , the concentrations of MDA were gradually de-creased and were lower than control significantly (P 〈 0.05 ). The percentage of TMS, PMI, NAR and mitochondrial membrane potential(Mt-MP) of the freezingthawing boar semen supplemented with M2 were significantly higher than those of control ( P 〈 0. 05 ) , and the concentration of MDA was lower significantly than control (P 〈 0. 05 ). In conclusion, addition of optimal concentra tion of MLT prevented spennatic oxidative damage from ROS and improved the quality of preserved boar semen.
