摘要
背景多巴胺通过D5受体对于机体的血压具有重要的调节作用。转人D5突变基因D5F173L小鼠的收缩压、舒张压和平均动脉血压均明显高于转人D5突变基因D5S390G及正常人D5基因(hD5WT)小鼠。目的应用超声心动仪观测人多巴胺D5受体突变基因F173 L(D5F173L),S390G(D5S390G)及正常人D5基因(hD5WT)的转基因小鼠心脏结构和功能的变化,探讨该基因对心脏疾病发病机制的影响。方法利用稳定传代的D5F173L,D5S390G及hD5WT基因转基因小鼠,通过超声心动仪观察各组转基因小鼠及阴性对照小鼠舒张期及收缩期左心室结构及功能的变化。结果超声心动仪结果显示自4~5月龄始,D5F173L转基因小鼠的舒张期与收缩期左心室容积与左心室内径均明显小于多巴胺hD5WT转基因小鼠及阴性对照小鼠(n=6,P<0.05),其舒张期与收缩期左心室前后壁厚度及左心室质量则明显高于多巴胺hD5WT转基因小鼠及阴性对照小鼠(n=6,P<0.05)。而D5S390G小鼠舒张期与收缩期左心室内体积、左心室内径、左心室前后壁厚度与多巴胺hD5WT转基因小鼠及阴性对照小鼠比较无显著差异。结论多巴胺D5转基因小鼠在血压升高的同时伴有心脏结构与功能的变化,提示多巴胺D5在心脏结构与功能形成过程中具有重要的调节作用。
Background Dopamine via D5 receptor plays an important role in regulating blood pressure. The systolic, diastolic and mean arterial blood pressures of human D5 mutated gene DSF1731. transgenic mice are bigher than the human D5 mutated gene D5S390G and human wild type D5, gene transgenic mice. Objective To explore the effeet of human dopamine D5 polymorphism genes D5F173L and D5S390G, and wild type human D5(hFSWT) gene in heart structure and function. Methods Investigation of systolic and diastolic structure and function in heart was used by ultrasongraphy. ResultsThe volume and inner diameter of left ventricule (LV) are smaller; the anteri or and posterior thickness of LV are thicker, and the weight of LV are heavier in D5F173L transgenic mice than hD5WT transgenic and non-transgenic mice since 4 5 months of age ( n=6, P〈 0.05 ). The changes are not found in D5S390G transgenic mice comparing with hD5WT transgenic and non-transgenic mice. Conclusion The results suggest that the dopamine D5 receptor plays an important role in the development of essential hypertension and the regulation of heart structure and function. However, the mechanisms remaiu to be studied.
