摘要
目的探讨老年大鼠肝脏枯否细胞的前体细胞即外周血单核细胞对内毒素的反应性。方法将不同浓度脂多糖(内毒素)加入经培养的6及24月龄大鼠单核细胞中,2小时后检测单核细胞细胞骨架中肌动蛋白、肌球蛋白、波形蛋白等成分的含量。结果老年大鼠单核细胞骨架成分的含量显著低于年轻大鼠(P<0.01)。低浓度脂多糖使单核细胞骨架成分的含量增加,以年轻大鼠更为显著(P<0.01);高浓度脂多糖使单核细胞骨架成分的含量减少,以老年大鼠更显著(P<0.01)。结论老年大鼠单核细胞细胞骨架老化明显,更易受内毒素的损伤。
Objective To evaluate the reactivity of monocytes, the precursors of Kuffer's cells, to endotoxin lipopolysaccharide in aged rats. Methods Lipopolysaccharide of various concentrations was added into the suspension of monocytes, and the contents of actin, myosin and vimentin in the cytoskeleton were measured two hours later. Results Cytoskeleton contents of aged rats were distinctly lower than those of young rats (P<001). Lipopolysaccharide of low concentration led to increase cytoskeleton contents,which were more prominant in young rats (P<001). Lipopolysaccharide of high concentration led to decrease cytoskeleton contents, which were more prominant in aged rats (P<001). Conclusions The results indicate that the cytoskeleton of monocytes in the aged rats becomes aged distinctly and is more easily injuried by endotoxin lipopolysaccharide.
