绿菜花双核期小孢子比例对游离小孢子培养的影响 被引量:21

Effect of Binucleate Pollen Ratio on Embryogenesis in Isolated Microspore Culture of Broccoli

摘要 在1/2NLN培养基、32.5℃、24h热激处理下,对3份绿菜花品种进行游离小孢子培养,结果表明,悬浮培养液中双核期小孢子比例对胚胎发生影响很大。品种的基因型不同,适宜的双核期小孢子比例也不同,‘bc-0’、‘Galaxy’在双核期小孢子比例分别为15.38%和13.60%时,出胚率最高;‘bc-4’则在50.7%时出胚率最高,达37.08胚/蕾。 Isolated microspores of 3 genotypes of broccoli were cultured in 1/ 2NLN medium, pretreated at 32.5℃ for 24h. Results showed that the binucleate pollen ratio affected embryogenesis significantly. Different genotypes had different proper binucleate pollen ratio. For ‘bc-0’ and ‘Galaxy’ , 15.38% and 13.60% binucleate pollen ratio gave the highest embryo yield; For ‘bc-4’ , the highest embryo yield was obtained with 50.7% binucleate pollen ratio and it was 37.08 embryo per bud.
