摘要
目的:探讨脂多糖(lipoppolysacchride,LPS)作用于人子宫内膜细胞引起的炎症相关基因表达谱的改变,为防治子宫异常出血提供靶基因。方法:收集增生期人正常子宫内膜3例进行传代培养,将收获的人子宫内膜细胞分LPS组与空白对照组,LPS组培养液中加入50μg/ml的LPS培养24h,应用基因芯片技术通过计算机扫描分析对照组和LPS组基因表达谱的差异情况。结果:与对照组比,LPS组出现表达差异的基因涉及很多方面的相关基因,其中与炎症相关的基因有IL-1β、IL-8等。结论:这些与炎症相关的基因可能与LPS诱导子宫异常出血有关。
Objective: To investigate differentially expressed genes related to inflammation in endometrial cells induced by lipopolysaccharide(LPS) in order to provide target gene for treating abnormal uterine bleeding. Methods: The endomentrial cells from 3 cases of normal endometrium in hyperplasia stage were divided randomly into LPS group and control group. In LPS group, 50 μg/ml LPS was added into the cultured solution and cultured for 24 h. The differences of gene expression profile was analyzed by laser scanner between the two groups. Results: A total of differentially expressed genes were found, these related to many aspects, moreover, among these, the expression of genes involved in inflammation were up-regulated by LPS, such as overexpression of IL-1β, -8, etc. Conclusion: Abnormal uterine bleeding induced by LPS is associated with genes related to inflammation.
