乙型肝炎肝纤维化组织中DDAH1的功能研究

摘要 目的研究慢性乙型肝炎肝纤维化组织中二甲基精氨酸二甲基氨基水解酶1(DDAH1)的表达,并初步探讨DDAH1在肝纤维化发生、发展中的作用。方法采用Western blot检测肝纤维化组织中DDAH1表达水平,进一步测定肝组织中DDAH1活性、不对称二甲基精氨酸(ADMA)含量,一氧化氮合酶(NOS)活性及NO2-/NO3-含量。结果肝纤维化组织中DDAH1表达下降、活性降低,ADMA含量增加,NOS活性下降,一氧化氮(NO)含量减少。结论DDAH1可能通过调节ADMA代谢,影响NO合成,从而参与了肝纤维化的发生和发展。 Objective To investigate the expressions of DDAH1 in hepatic fihrotic tissues of patients with chronic hepatitis B, and analyze the functions of DDAHI in the formation and development of hepatic fibrosis. Methods The expression of DDAH1 was analyzed by Western-blot in hepatic fibrosis tissues and the normal liver tissues. DDAH's activity, ADMA concentration, NOS activity and NO concentration are to be determined. Results The expression of DDAH1 by western blot showed decrease, and further study showed that the activity of DDAHI was lowered with the increase of ADMAin hepatic fibrosis tissues [(2.04±0.33)/amol/L vs (1. 16 ±0. 21)/μmol/L, P〈0.01], decreasing activity of NOS [(4.39± 0.66) U/ml vs (7.65 ±0.91) U/ml, P〈0.05] and decrease of concentration of NO decrease (55.72 ±11. 32 vs 96.27 ± 17.84, P〈0.01). Conclusion DDAHI may involve in the formation and devel.opment of hepatic fibrosis by regulating ADMA metabolism and impacting NO synthesis.
